nature
10-desacetylbacatin III(10-DAB III) is a tetracyclic diterpenoid natural taxane compound extracted from the branches and leaves of Taxus, which is a white to off-white crystalline powder at room temperature. It has a melting point in the range of 231-236°C and a specific rotation [α] D²³ of − 44.5 °(c = 1, methanol). Solubility, the product is soluble in methanol (heat), ethanol, dichloromethane, DMSO, chloroform and other organic solvents, slightly soluble in acetonitrile, almost insoluble in water. The molecular structure contains a number of hydroxyl and ester groups, with a strong hydrogen bond donor/acceptor capacity (4 hydrogen bond donor, 10 hydrogen bond acceptor), lipid water partition coefficient LogP is about 0.6, showing moderate polarity. Chemically, the C- 13-position hydroxyl group has high reactivity and is the key reaction site for semi-synthetic paclitaxel and docetaxel; the C- 10-position is the free hydroxyl group after deacetylation, and its activity is significantly different from that of bacatin III. The product is sensitive to light, heat and moisture, and should be stored under dark, dry and low temperature conditions to prevent degradation.
Use
10-DAB III is the core intermediate in the semi-synthetic system of taxanes. It is a precursor product for the synthesis of paclitaxel (Paclitaxel) and docetaxel (Docetaxel, also known as docetaxel), and is also used in the preparation of other paclitaxel analogs. Paclitaxel and docetaxel are both widely used anti-cancer drugs in the world, and 10-DAB III can be extracted from the branches and leaves of yew, without cutting down the whole plant, so it has important industrial value in terms of sustainable supply and cost control. In addition, the study also showed that 10-DAB III itself has a certain inhibitory effect on Leishmania promastigotes in vitro, suggesting that it also has room for exploration in the field of anti-parasites.
Safety Information
according to GHS classification information, 10-DAB III has a hazardous material designation of T (Toxic Substances) and a hazard category code of R23/24/25, indicating toxicity by inhalation, skin contact and ingestion. Wear protective clothing, protective gloves and goggles/face shield during operation; use respiratory protective equipment in poorly ventilated areas; in case of accident or discomfort, seek medical attention immediately and show the product container label. The transport number of dangerous goods is UN 1544, the hazard class is 6.1 (B), and the packing class is III. Store away from fire and oxidizing agents, keep the container sealed, and place it in a dry environment at 2-8°C in the absence of light. This product is for research and development and production purposes only, and cannot be used in humans or as a household backup medicine.
